好的,各位朋友大家好,欢迎收听本期的《科学播报》。口腔溃疡总是影响我们的日常饮食,让人浑身难受。但是仔细回想一下,似乎在同等情况下,相对于其它地方的皮肤伤口而言,口腔里的伤口愈合好像速度更快,也更不容易形成伤疤。这种感觉是真的吗?具体机理又是什么呢?
近日,美国国立关节肌肉骨骼及皮肤病研究所和加州大学圣迭戈分校的研究人员对比了口腔和皮肤伤口的愈合情况并分析了愈合过程中分子表达谱,发现口腔中伤口的愈合速度确实要比皮肤伤口更快,而这与两种转录因子息息相关。它们的表达在口腔粘膜出现伤口后持续上调,控制上皮细胞分化并调节炎症反应,使得口腔伤口更快速地愈合。
研究人员组织了一批健康的志愿者,在他们同意的前提下在口腔里和手臂皮肤上分别划两个3 mm和5 mm的小伤口。在15天内,定期检查志愿者的皮肤和口腔伤口,并做连续活检。实验结果正如预期的那样,口腔里的伤口愈合速度明显快于皮肤伤口。比如,口腔伤口为3 mm时平均的愈合时间约6天,而同样状态下的皮肤伤口大约需要两周。更重要的是,研究人员发现两处细胞的基因表达情况有明显不同。
研究人员发现,同一种基因,在皮肤伤口愈合过程中表达活性明显增强,而在口腔伤口中表达活性增强较小。该发现证明在口腔粘膜上皮细胞中,未受伤时就已经存在受伤激活的转录体系,也就是说,即使没有受伤,口腔粘膜也一直在为伤口愈合做准备。在伤口愈合过程中,口腔粘膜上皮细胞中的一些基因活性比皮肤细胞的高,这些基因所编码的转录因子能够促进细胞迁移和抑制炎症反应,有利于促进愈合并减少疤痕。
这项工作揭示了口腔伤口愈合快和疤痕少的分子机理,有助于开发慢性和难愈合性伤口的治疗方案。而至于为什么会有这样的现象,研究人员认为,人类,作为从海洋中进化而来的脊椎动物,我们的祖先已经习惯了在水下愈合伤口,而口腔和各种粘膜所处的环境,又与海洋环境极为相似,祖先们遗留下来的基因使得口腔中的伤口在这样的条件下愈合得更快。
