生命之树的基础分为三个领域。其中一种被细菌占据——没有细胞核或膜结合细胞器的单细胞微生物,并通过挥舞称为鞭毛的发状结构而运动。另一个是真核生物,其细胞具有细胞核和膜。该域包括我们人类,动物,植物和藻类。还有一种是古细菌。古细菌很像细菌,因为它们同样缺乏细胞核和与细胞膜结合的细胞器,并且可以利用鞭毛四处游荡。但是有一些关键差异,它们的细胞壁和RNA差异足以将它们分离到不同的生命枝丫上。
早在一百多年前,就诞生了一种被称为内共生学说的生命起源假说,该学说认为,真核细胞中的线粒体和叶绿体等细胞器,起源于共生于真核生物细胞中(之内)的原核生物。线粒体起源于好氧性细菌,而叶绿体源于内共生的光合自养原核生物的蓝细菌。
新的研究表明,该假说可能进化为一个更激进的解释。今年,微生物学家朝着解决关于真核生物起源的争议迈出了重要的一步。新的研究结果可能会将生命领域从三种(古细菌,真核生物和细菌)缩小到两种:细菌和古细菌,而真核生物被简化为古细菌的一个子集。但是鉴于证据不足,许多研究人员对此表示怀疑。

许多科学家认为,阿斯加德(Asgard)古细菌的祖先带来了真核生物的生命。大约20亿年前,阿斯加德的亲缘生物将细菌包裹起来,后来又演变成线粒体。
早在2010年,当科学家对北冰洋海底沉积物核进行DNA分析时,他们就有了令人惊讶的发现。先前未知的属于古怪微生物领域的生物称为古细菌,它们似乎具有与完全不同的领域——真核生物相关的基因组特征。
2015年,埃特玛(Ettema)和他的同事们在格陵兰岛沿海海底热液喷口场洛基城堡附近的泥浆中发现了神秘微生物的样本。他们回收了散布有真核生物样基因的古细菌基因组,不久之后,其他实验室发现了类似的古细菌。
直到现在,科学家们还是从这些样本的孤立遗传片段中整理谱系。

经过12年的尝试,日本的一个研究小组成功地从深海沉积物中培育出了神秘微生物,并对其基因组进行了测序。这种生物为Prometheoarchaeum syntrophicum菌株MK-D1,是最近被认可的阿斯加德微生物群的成员,这些微生物不是细菌,而是一种完全独立的生命分支,称为古细菌。
这种奇怪的生物最初是从日本沿海海拔2500米深的Omine Ridge中提取,被埋在深海淤泥中。为了模拟海床的恶劣条件,日本海洋地球科学技术局的微生物学家们建造了一个生物反应器来容纳海底泥浆,并不断将其浸入甲烷气体中。五年后,他们将样品刮入装有各种营养物质的玻璃管中,静待其萌芽。

一年后,他们发现其中一个样本表面上长有细菌,但在遗传上却截然不同,其中的微生物却在生长。这些是他们所希望的阿斯加德古细菌。在接下来的六年中,他们致力于分离和培养纯生物样品,他们发现需要14到25天才能使培养的细胞倍增。相比之下,大多数细菌在一小时内就会翻倍。
研究人员选择用普罗米修斯(Prometheus)命名,是因为这位希腊神从泥浆中雕琢了人类。
令人惊讶的是,这些片段包含的基因以前被认为仅在真核生物中发现,而真核生物是具有细胞核和细胞器的细胞,如线粒体。比较DNA分析表明,阿斯加德或其古老的亲缘生物甚至可能产生了真核生物。
研究还发现,微生物似乎与某些细菌结合在一起生长最好,并且它形成的短触手可能会吞噬细菌伴侣。如果是这样,那可以解释它如何获得成为线粒体的微生物。
今年的其他研究已从研究组其他成员的DNA片段中鉴定出更多的真核基因。来自DNA的有关阿斯加德代谢的信息也支持二域假说,而不是三域。支持这两种观点的人都同意,但是,将近30亿年前发生的事件将难以重构,而且随着对阿斯加德的研究,新的想法也可能会出现。但是现在有了研究对象,研究人员可以更清晰地了解遥远的过去与生命的起源。
