  多年以来,科学家一直想要破解精神疾病的致病机制。至今为止,人们对于多种精神疾病,包括自闭、精神分裂、抑郁等,仍然知之甚少。直到本世纪初,德国科学家在有关患者的遗体中有了新发现,研究才得到了新的进展。依据当时的分析结果,在精神疾病患者的大脑内,其小胶质细胞的活性异于常人。

  小胶质细胞是神经胶质细胞的一种,也是特殊的巨噬细胞。在大脑早期发育过程中,会有部分神经元发生凋亡等待清除。小胶质细胞的主要作用就是参与这些神经元的清理过程。另外,它也有吞噬病原体和清除受损神经元的作用。

  小胶质细胞与免疫系统密切相关。小胶质细胞出现异常,就会破坏人体免疫系统的正常工作。小胶质细胞又与神经元相关,而精神疾病属于神经系统疾病。因此科学家认为,精神疾病可能与免疫系统的异常有关。

  过去一系列的研究表明,在妊娠时期,如果胎儿的免疫系统受到病毒之类的感染破坏,就很有可能导致后来神经系统的发育失常,引发精神疾病。在胎儿发育过程中,小胶质细胞会协助将不必要的神经网络去除。而出现异常的小胶质细胞,就很可能对这个过程形成严重的干扰。

  为了进一步研究小胶质细胞与精神疾病之间的联系,研究人员通过小鼠进行更加深入的观察。研究人员发现,小鼠的小胶质细胞在成形的阶段十分敏感。小胶质细胞会经历早期、成形和成熟三个发育阶段。研究人员通过对不同阶段细胞的测试,确定小胶质细胞在成形阶段,一旦受到外部破坏性的干扰,相关的基因就会提前进行表达,使其活性异常增强。

  活性增强的小胶质细胞会错误地清除神经系统中必要的部分,使小鼠大脑的一些功能出现异常。这些异常的小鼠在出生后,表现出的行为与精神分裂、交流障碍等患者的行为十分相似。这些结果给了研究人员诸多启发,小胶质细胞与精神疾病的发生可能存在直接的联系。

  过去各种对于自闭、抑郁、精神分裂等症状的研究总是进展缓慢。其中的一个原因在于,要在技术层面上对患者的大脑进行深入研究十分困难。不过随着科技发展,现今科学家们可以利用新的手段,对精神疾病患者的大脑进行分析。

  例如最近,日本科学家通过阳离子断层成像技术(PET)直接对患者大脑进行观察,进一步发展了多年以来的猜想。这种成像技术对人体中的物质进行标记,进而通过显像仪器观察其聚集状态、变化过程。科学家发现,精神疾病患者的大脑中,小胶质细胞确实存在过度活化的现象。

  在不同年龄阶段的大脑中,小胶质细胞会表现出各不相同的活性。而在不同类型的精神疾病患者大脑中,小胶质细胞则表现出了各不相同的活性异常现象。因此科学家认为,破解小胶质细胞的不同异常所对应的不同精神疾病症状,正是破解精神疾病致病机制的一个关键所在。所有这些新的发现,都将为研究者未来开发新药物和治疗有关患者起到至关重要的作用。
